Restriction digests of the clone give the following sizes (kb): PstI--5.3, 4.1, 3.75; EcoRI--7.7, 3.9, 2.45; BamHI--12.0, 1.55. The order of fragments is as follows: PstI-BamHI (vaccinia)--2 kb; BamHI-BamHI (influenza)--1.78 kb; BamHI-PstI (vaccinia)--3.5 kb. The 2.0 kb BamHI fragment also contains an AvaI site and an EcoRI site. This was constructed by inserting a 5.5 kb PstI fragment from vaccinia into pBR325 (modified to remove the BamHI site). A 1.8 kb HindIII fragment encoding hemagglutinin was linkered with BamHI and inserted into a BamHI site of the vaccinia insert. |
