The clone contains the following restriction sites (using the viral numbering scheme from 1 to the SalI site at 4833): BamHI--1926, 2148, 8921; EcoRI--408, 2414, 2474, 8546, 10009, 10681; SacI--3750, 10185; PvuII--4000, 6249, 10681; SalI--4833, 9196. Restriction digests of the clone give the following sizes (kb): SalI--8.2, 4.4; EcoRI--6.5, 2.5, 2.2, 1.5, 0.5; HindIII--4.3, 4.0, 1.2, 1.0, 0.6 (doublet), 0.5; BamHI--6.8, 6.0; PstI--6.5, 4.4, 2.2. Known genes have these approximate positions (virus numbering scheme): I: 300-1300; II (aphid helper component): 1300-1800; III: 1800-2200; IV (coat protein):2200-3600; V (reverse transcriptase): 3600-5600; VI (inclusion body protein): 5800-7300. This strain was isolated from crucifers in Davis, California. This was constructed by linearizing the full-length CaMV DNA with SalI and inserting it into the SalI site of pBR322. |
