The clone contains the following restriction sites (using the viral numbering scheme from 1 to the SalI site at 4833): BamHI--1926, 2148, 5108; EcoRI--2414, 2474, 5483, 10009, 10468; SacI--10185; PvuII--4000, 7780, 10681; SalI--4833, 9196. Restriction digests of the clone give the following sizes (kb): SalI--8.0, 4.3; EcoRI--4.3, 4.2, 3.8, 0.4; HindIII--4.1, 3.8, 1.1, 1.0, 0.6 (triplet) 0.3; PstI--8.4, 3.9. Known genes have these approximate position (virus numbering scheme): I: 300-1300; II (aphid helper component): 1300-1800; III: 1800-2200; IV (coat protein): 2200-3600; V (reverse transcriptase): 3600-5600; VI (inclusion body protein): 5800-7300. This strain was isolated from crucifers in the Mendoza province of Argentina. This was constructed by linearizing the full-length CaMV DNA with SalI and inserting it into the SalI site of pBR322. |
