Restriction digests of the clone give the following sizes (kb): HindIII/SalI--3.1, 1.65, 1.3, 0.75, 0.5 + smaller; XbaI--4.4, 3.4; EcoRI--7.8; SmaI--4.3, 3.5; SphI--7.8. This is an integrative, selectable vector conferring arginine prototrophy on arginine autotrophic hosts such as Aspergillus niger 350(-)52, ATCC 20739 (argB-). It also confers kanamycin resistance. This was constructed by inserting a HindIII/SalI fragment of Tn5 into pUC12 cut with HindIII/SalI (pNE02) and then inserting an XbaI fragment from pBB116 into the XbaI site of pNE02. |
